Some Techniques in Mycology
Riddell's slide culture technique. Because Mycologists rely so heavily on the microscopic morphology of fungi to identify them it is essential to have a good microscopic preparation. One has to observe the precise arrangement of the conidiophores and the way in which the conidia are produced (conidial ontogeny). Riddell's simple method of slide culture (Mycologia 42: 265, 1950) permits fungi to be studied virtually in situ with little disturbance.
Specimen collection tools for Superficial Mycoses
Some of the common instruments used for the collection of Skin, Nail and Hair specimens for diagnosis of superifical mycotic agents.
WOOD LAMP
Wood's ultra-violet light. Note simulated greenish-yellow fluorescence in small beaker. This device is useful as a presumptive diagnosis in the management of superficial cutaneous fungal infections.
Golden yellow fluorescence - Malassezia furfur..
Bright Green - Microsporum canis, Microsporum audounii.
Dull Green - Trichophyton schoenleinii.
RACE TUBES
These elonagated tube are employed to study the circadian rhythms in rapid growing fungi.
INVITRO METHOD FOR ESTIMATION OF SAP ACTIVITY
Note the Halo indicative of the Proteolytic effect around the Colony of Yeast (ATCC 24433 - C. albicans) after Amido Black Staining ( Staib etal. 1965)
Circadian rhythm exhibited by Aspergillus niger
